Does the formula for a point load (PL/4) on beams change if the load is not acting on the mid span of a beam?

PL/4 is the formula (or ‘equation’) for the maximum bending Moment of a simple span beam with a point load at mid-span. Yes, the formula changes when the point load is not at mid-span. The maximum moment decreases as the point load moves away from mid-span. The formula becomes Pab/L where ‘a’ and ‘b’ are the distances to the point load from the left and right ends, respectively.
